H2O2 Participates in the Induction and Formation of Potato Tubers by Activating Tuberization-Related Signal Transduction Pathways

Reactive oxygen species (ROS), especially hydrogen peroxide, H2O2, act as signaling molecules to widely mediate growth, development, and stress response of plants. In the present study, internal ROS accumulation, effects exogenous H2O2 treatment, expression key tuberization-related genes, effect knockout Solanum tuberosum self-pruning 6A (StSP6A) on H2O2-induced tuber formation were investigated elucidate whether how is involved in induction potato tubers using two diploid landraces, phureja S. ajanhuiri. The results showed that there was a significant accumulation (including superoxide anion, O2−, total ROS) during stolons/tubers, hook-like subapical part stolons prior tuberization, detected by staining observation quantitative measurement. Furthermore, treatment significantly enhanced percentage formation. By contrast, addition either inhibitor diphenyleneiodonium chloride (DPI) or scavenger catalase (CAT) resulted decline addition, analysis nine genes demonstrated tuberization could be associated with H2O2-controlled regulation these tuberization- signaling-pathway-related StSP6A, which dramatically up-regulated early stage treatment. When StSP6A knocked out CRISPR-Cas9-mediated genome editing, frequency null-mutants became lower at various concentration treatments. These findings indicate might play an important role acting molecule initiate induction, triggered regulating gene activating signal transduction pathways, pivotal player potato.
